Question
How gene therapy is given?

Answer

→There are two main approaches of gene therapy.
(1) Ex vivo means out side of the body in which the cells are removed from the patient's blood and or bone marrow and culture them in the laboratory.
→ In this cells, desirable gene containing virus can be introduced.
→ When viruses enter into the cell, the desired DNA becomes the part of genome.
→These cells are grown in the laboratory and then injected to the veins of patient.
(2) In vivo means inside the body, no cells are removed from the patient's body, but by using vectors desirable gene is introduced into the body of patient.

A relevant portion of B-chain of haemoglobin of a normal human is given below:

The codon for the sixth amino acid is GAG. The sixth codon GAG mutates to GAA as a result of mutation 'A' and to GUG as a result of mutation 'B'. Haemoglobin structure did not change as a result of mutation 'A', whereas haemoglobin structure changed because of mutation 'B', leading to sickle-shaped RBCs. Explain giving reasons how could mutation 'B' change the haemoglobin structure or bring down mutation and not mutation 'A'.
