Question
What is micro-injection? How it is helpful in recombinant DNA technology?
✓
Answer
Micro-injection : In this method, recombinant DNA is directly injected in the nucleus of animal cell. For this purpose a glass pipette is used whose diameter is 0.1 - 0.5 u. For this process, well equipped laboratory instruments are required. However, with the help of these tools, DNA can be injected within 1 Hour in approx. 500 - 1000 cells. About 50% of the injected cells express the injected DNA. It has been observed that the injected DNA will randomly attached within nuclear DNA and will express itself after finding suitable promoter. Characteristic feature of this method is that may fragment of DNA can be injected in any cell. Any selectable force is not required for maintenance of translated gane. This technique has been used in rats by transferring growth hormone genes. Some of the genes express and give rise giant sized rats. The similar technique is used in plants also. There are some disadvantages of this technique, as it is very expensive and requires skill perfection.
