Origin of Replication = ori: This is the sequence from where replication begins and when a segment of any DNA gets attached to this sequence then it can replicate inside the host cells. This sequence is also responsible for controlling the number of copies of DNA added. Therefore, if one wants to obtain a large number of target DNA, it should be cloned into a vector whose origin (ori) supports high copy number.
